Do the big clubs use and more importantly inforce NDA's on all their staff? (Non Disclosure Agreements)

YES.

Every organisation use them if they see fit and this is quite normal business practice

It seems like agents use NDA's to wipe their cracks with though, so I guess they aren't bound by them often

They wouldn't have NDA's with staff, they would have confidentiality clauses in their employment contracts. Typically NDA's are used where no other contract is in place.
